Could herpes virus play a role in Alzheimer's?

A new study suggests that certain viruses could kick-start an immune response that might increase the accumulation of amyloid, a protein in human brains which clumps into the telltale plaques of Alzheimer's.

Herpes viruses HHV 6A and 7 were found in greater abundance in brain samples from AD patients compared with normal brains. After an 18-month study examining the effects of nutritional compounds found in foods such as trout, broccoli and peppers on people with Alzheimers, the team from the Nutrition Research Centre Ireland (NRCI) claims to have done a ‘statistically significant finding.

Once the researchers realised that these viruses have a link to the Alzheimer's disease, they immediately tried to identify what role the two herpes virus genes played in affecting the other genes in the brain.

They found a lot of interactions, suggesting the viruses could even switch on and off Alzheimer's-related genes.

However, since the strains were discovered in both Alzheimer's brain tissue and non-Alzheimer's brain tissue, it is quite hard for the researchers to ascertain that Alzheimer's disease is primarily caused by the viruses.




However, others were more sceptical. By mid-century, a new diagnosis will occur every 33 seconds, and costs are expected to exceed $1 trillion annually. From as early as the 1950s, people have been making assumptions that pathogens could be the main contributors of the disease.

There have been earlier studies hypothesizing that viral infections could be the key to Alzheimer's disease. The combined funding support for this five-year endeavor is $185.2 million. But with the current research's findings, researchers may one day be able to develop antiviral Alzheimer's treatments.

However, Joel Dudley, from the Icahn School of Medicine in NYC and the lead author of the city said that just having the two herpes viruses in the brain is not enough to cause the Alzheimer's disease. The data they mined is usually discarded, but was archived instead by the National Institutes of Health in a bid to accelerate the discovery of new treatments by fostering "big data" collaborations. Dr. Tanzi and his Harvard colleague Dr. Robert Moir (unrelated to this study) looked at the sticky beta-amyloid captures that invaded germs and causing the forming of plaque. The authors emphasize that their findings do not prove that the viruses cause the onset or progression of Alzheimer's. What's more, people with ApoE4, the gene known to increase one's risk of Alzheimer's, had especially high levels of HHV-6, as study author Ben Readhead told The New York Times.

Readhead said that there is a need for more research to determine the extent to which the viruses are involved in causing the disease. The research team assessed up to 1000 postmortem brains of people who had Alzheimer's disease and those who did not. Dudley is a member of the ASU-Banner Neurodegenerative Disease Research Center.
